The project was jointly declared by the Suzhou Institute of Biomedical Engineering and Technology and the Institute of Biophysics of the Chinese Academy of Sciences in 2014. Among them, the Suzhou Institute of Medical Technology is a research and development unit, and the Institute of Biophysics is a user unit. The research and development work was carried out by the research group Li Hui of the Suzhou Institute of Medical Sciences. In September 2016, the non-linear SIM super-resolution microscope developed by the Li Hui team was sent to the Institute of Biophysics for trial and trial. In this system, the research team proposed a super-resolution imaging method based on structured light activation + structured light excitation for low light nonlinear structure light illumination, and replaced the mechanical grating with a ferroelectric liquid crystal spatial light modulator, in parallel with the FPGA parallel synchronous control system. , to achieve a more flexible imaging and faster imaging speed. At the same time, the research group developed a SIM/NL-SIM super-resolution image reconstruction algorithm and software that can be applied to weak signal samples. Using this device, biological samples such as fluorescent microspheres, endoplasmic reticulum, mitochondria, cell nuclei, and cytoskeleton were observed, achieving lateral resolution of 100 nm in linear SIM mode and 62 nm in non-linear SIM mode.
The expert group listened to the project work report, financial report, user use report, and conducted field test and acceptance. After on-site testing and full discussion, the expert group believes that the technical indicators of the project meet or exceed the requirements of the implementation plan and meet the requirements of super-resolution observation of biomedical imaging applications.
